Application & Usage Western blotting (0.5-4 µg/ml). However, the optimal concentrations should be determined individually. The antibody recognizes mainly ~16 kDa band in human, mouse and rat samples. A ~34 kDa band is also detected to the lesser extent. Reactivity to other species has not been tested.

Function Promotes activation of caspases and apoptosis. Promotes mitochondrial membrane changes and efflux of apoptogenic proteins from the mitochondria. Contributes to p53/TP53-dependent apoptosis after radiation exposure. Promotes proteasomal degradation of MCL1. Competes with BAK1 for binding to MCL1 and can displace BAK1 from its binding site on MCL1 (By similarity). Competes with BIM/BCL2L11 for binding to MCL1 and can displace BIM/BCL2L11 from its binding site on MCL1.

Background

Noxa is a BH3-only member of the Bcl-2 family. Expression of Noxa gene involves direct activation of its promotor by p53. Increased expression of Noxa protein occurs in normal thymocytes but not in p53-deficient thymocytes. Coimmunoprecipitation data s µggest that Noxa protein may interact with proteins belonging to the Bcl-2 family, such as Bcl-xL and Mcl-1. Blocking the endogenous Noxa induction results in the suppression of apoptosis. These s µggest Noxa may represent a candidate mediator of p53-induced apoptosis.
