| Function | G protein-coupled receptor for melanocyte-stimulating hormones (alpha- beta- and gamma-MSH) and corticotropin/ACTH, which are peptide products of the POMC precursor (PubMed:37524700, PubMed:8396929). Upon activation, couples to G(s) protein, stimulating adenylate cyclase and the cAMP-dependent signaling pathway (PubMed:37524700). Also activates ERK1/2 via a PI3K-dependent signaling mechanism (PubMed:19428994). Order of potency of natural melanocortins in receptor activation is alpha-MSH > ACTH > beta-MSH > gamma-MSH (PubMed:8396929). Plays a key role in immune response, and is essential for temperature regulation and exocrine gland function (By similarity). |

Background
Receptor for MSH (alpha, beta and gamma) and ACTH. The activity of this receptor is mediated by G proteins which activate adenylate cyclase. This receptor is a possible mediator of the immunomodulation properties of melanocortins.
