Application Note | This antibody has been tested for use in immunofluorescence microscopy and western blotting. Specific conditions for reactivity should be optimized by the end user. Expect a band approximately 13 kDa in size corresponding to mature MHV-A59 nsp9 by western blotting in the appropriate cell lysate or extract. For immunofluorescence microscopy, Vero-E6 cells were grown on glass slides followed by infection with MHV-A59 strain and fixation with PBS/3% PFA. After washing and permeabilization of the fixed cells, antibody incubation was performed in PBS/5% FCS for 30 min. |
Physical State | Liquid (sterile filtered) |
Buffer | 0.02 M Potassium Phosphate, 0.15 M Sodium Chloride, pH 7.2 |
Immunogen | This antibody was produced in mice by repeated immunizations with E.coli derived full-length MHV-A59 nsp9 protein. This protein is part of the viral replicase polyprotein. |
Preservative | 0.01% (w/v) Sodium Azide |

Purity | This antibody is directed against the MHV-A59 nsp9 protein. This product was purified from tissue culture supernatant fluid by Protein A chromatography. No cross reactivity occurs with SARS CoV nsp9. Cross reactivity with homologues from other sources has not been tested. |
Storage Condition | Store vial at -20° C prior to opening. Aliquot contents and freeze at -20° C or below for extended storage. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4° C as an undiluted liquid. Dilute only prior to immediate use. |
Precautions Note | This product is for research use only and is not intended for therapeutic or diagnostic applications. |

Function | The papain-like proteinase 1 (PL1-PRO) and papain-like proteinase 2 (PL2-PRO) are responsible for the cleavages located at the N-terminus of the replicase polyprotein. In addition, PLP2 possesses a deubiquitinating/deISGylating activity and processes both 'Lys-48'- and 'Lys-63'-linked polyubiquitin chains from cellular substrates. Antagonizes innate immune induction of type I interferon by blocking the phosphorylation, dimerization and subsequent nuclear translocation of host IRF-3 (By similarity). |

Note=nsp7, nsp8, nsp9 and nsp10 are localized in cytoplasmic foci, largely perinuclear. Background
The nonstructural protein 9 (nsp9) is one of the Mouse hepatitis virus replicase cleavage products, encoded by ORF1a. Nsp9 is an RNA-binding protein that is thought to be part of the viral replication complex, which is associated with intracellular membranes.
