Application & Usage | Western blotting (0.5-4 µg/ml). Based on researchers feedback, it also works well in immunoprecipitation (10-20 µg/ml) and Immunohistochemistry (15-25 µg/ml). However, the optimal concentrations should be determined individually. The antibody recognizes 34 kDa TRADD in samples from human, mouse, and rat origins. Reactivity to other species has not been tested. Mouse small intestine tissue lysate can be used as a positive control. |

Function | Adapter molecule for TNFRSF1A/TNFR1 that specifically associates with the cytoplasmic domain of activated TNFRSF1A/TNFR1 mediating its interaction with FADD (PubMed:7758105, PubMed:8612133, PubMed:23955153). Overexpression of TRADD leads to two major TNF- induced responses, apoptosis and activation of NF-kappa-B (PubMed:7758105, PubMed:8612133). The nuclear form acts as a tumor suppressor by preventing ubiquitination and degradation of isoform p19ARF/ARF of CDKN2A by TRIP12: acts by interacting with TRIP12, leading to disrupt interaction between TRIP12 and isoform p19ARF/ARF of CDKN2A (By similarity). |

Background
TRADD (TNF-R1-associated death domain) is a 34 kDa protein that associates with the death domains of both FAS and TNF-R1 via their DDH regions. Overexpression of TRADD leads to NFκB activation and apoptosis in the absence of TNF.
