KD-Validated Anti-MCM6 Rabbit Monoclonal Antibody

Rabbit monoclonal antibody

  • WB - KD-Validated Anti-MCM6 Rabbit Monoclonal Antibody AGI1019
Western blotting analysis using anti-MCM6 antibody (Cat#61136). Total cell lysates (30 µg) from various cell lines were loaded and separated by SDS-PAGE. The blot was incubated with anti-MCM6 antibody (Cat#61136, 1:5,000) and HRP-conjugated goat anti-rabbit secondary antibody (Cat#201, 1:20,000) respectively. Image was developed using FeQ™ ECL Substrate Kit (Cat#226).

  • WB - KD-Validated Anti-MCM6 Rabbit Monoclonal Antibody AGI1019
Western blotting analysis using anti-MCM6 antibody (Cat#61136). MCM6 expression in wild type (WT) and MCM6 shRNA knockdown (KD) HeLa cells with 30 µg of total cell lysates. Hsp90 α serves as a loading control. The blot was incubated with anti-MCM6 antibody (Cat#61136, 1:5,000) and HRP-conjugated goat anti-rabbit secondary antibody (Cat#201, 1:20,000) respectively. Image was developed using FeQ™ ECL Substrate Kit (Cat#226).

  • FC - KD-Validated Anti-MCM6 Rabbit Monoclonal Antibody AGI1019
Flow cytometric analysis of MCM6 expression in C2C12 cells using MCM6 antibody (Cat#61136, 1:2,000). Green, isotype control; red, MCM6.

  • ICC - KD-Validated Anti-MCM6 Rabbit Monoclonal Antibody AGI1019
Immunocytochemical staining of C2C12 cells with MCM6 antibody (Cat#61136, 1:1,000). Nuclei were stained blue with DAPI; MCM6 was stained magenta with Alexa Fluor® 647. Images were taken using Leica stellaris 5. Protein abundance based on laser Intensity and smart gain: Medium. Scale bar: 20 µm.

Protein Information
Name MCM6 (HGNC:6949)
Function Acts as a component of the MCM2-7 complex (MCM complex) which is the replicative helicase essential for 'once per cell cycle' DNA replication initiation and elongation in eukaryotic cells. Core component of CDC45-MCM-GINS (CMG) helicase, the molecular machine that unwinds template DNA during replication, and around which the replisome is built (PubMed:16899510, PubMed:32453425, PubMed:34694004, PubMed:34700328, PubMed:35585232, PubMed:9305914). The active ATPase sites in the MCM2-7 ring are formed through the interaction surfaces of two neighboring subunits such that a critical structure of a conserved arginine finger motif is provided in trans relative to the ATP-binding site of the Walker A box of the adjacent subunit. The six ATPase active sites, however, are likely to contribute differentially to the complex helicase activity (PubMed:32453425).
Cellular Location Nucleus. Chromosome. Note=Binds to chromatin during G1 and detaches from it during S phase.
