    Rockland Goat anti Choline Oxidase antibody was used to detect Choline Oxidase. Samples of ~1 and 0.25 µg of purified Choline Oxidase per lane were run by SDS-PAGE under reducing and non-reducing conditions and reduced samples of purified Choline Oxidase contained 4% BME and were boiled for 5 minutes. Protein was transferred to nitrocellulose and probed with Rockland Goat anti-Choline Oxidase (p/n 200-101-094, lot 5911) at 1:5K in (p/n MB-070) ON at 4°C. Primary antibody was detected with Rockland Dylight 649 conjugated Donkey anti-Goat (p/n 605-743-125) at 1:10K for 1.5 hr RT in (p/n MB-070) and imaged on the BioRad VersaDoc imaging system.

Application Note Goat anti-Choline Oxidase Antibody has been tested by western blot and is suitable to be assayed against 1.0 ug of Choline Oxidase [Alcaligenes species] in a standard ELISA using Peroxidase conjugated Affinity Purified anti-Goat IgG [H&L] (Rabbit) code #605-4302and (ABTS (2,2’-azino-bis-[3-ethylbenthiazoline-6-sulfonic acid]) code # ABTS-100 as a substrate for 30 minutes at room temperature.  A working dilution of 1:4,000 to 1:32,000 of the reconstitution concentration is suggested for this product.

Additional Information
Purity This product was prepared from monospecific antiserum by a delipidation and defibrination. Assay by immunoelectrophoresis resulted in a single precipitin arc against anti-goat serum, purified and partially purified Choline Oxidase [Alcaligenes species].  Cross reactivity against Choline Oxidase from other sources is unknown.
Storage Condition Store vial at 4° C prior to restoration.   For extended storage aliquot contents and freeze at -20° C or below.  Avoid cycles of freezing and thawing.  Centrifuge product if not completely clear after standing at room temperature.  This product is stable for several weeks at 4° C as an undiluted liquid.  Dilute only prior to immediate use.

Protein Information
Name CHOX
Function Oxidizes choline to betaine, probably via two successive reactions with betaine aldehyde as an intermediate (PubMed:6997283). Accepts either choline or the reaction intermediate betaine-aldehyde as substrate (PubMed:6997283). Glycine-betaine accumulates to high levels in the cytoplasm of cells to prevent dehydration and plasmolysis in adverse hyperosmotic environments (By similarity).

Choline Oxidase catalytic activity is Choline + 2 O2 + H2O = betaine + 2 H2O2. This protein is involved in step 1 of the subpathway that synthesizes betaine from choline.
